Can somebody suggests the various techniques that can be used to analyse the deaminated peptide.

I assume you meant to write "deamidated." The conversion of glutamine to glutamate or asparagine to aspartate changes the net charge. Therefore the most common methods are weak anion exchange or weak cation exchange. The variant will elute early by cation exchange or late by anion exchange. If the peptide is not too big, typical reversed-phase conditions for peptide analysis (H2O/MeCN/TFA gradient) will also do the job. Of course, you can combine the separation modes.

Is there any way to confirm the absence of deamidated peptide in the API. I donot have any deamidated peptide standard available with me.
