I need to know how "build up" a good method of hplc analisys of Doxazosin Mesylate in human serum (et/or urine).......

Please help me... (I'm so ignorant....... :shock: )

The more difficult part is not the HPLC method, but the sample cleanup.

For the HPLC method, I suggest to run a gradient from 5 to 95% organic on a 5 cm x 4.6 mm 3 micron column over 20 to 30 mL of gradient volume. Initial pH set up with 20 mM acetate buffer on a modern C18 column.

For the sample preparation, I definitely recommend solid-phase extraction. Look at the Waters website for the Oasis applications notebook! It contains detailed information how to do this.
