Hi,

We tried to determine the residual imidazole content by HPLC technique by using the ion pair mobile phase. We are geting the imidazole elutiion, but always peak shape is not good. By changing the diluent the peak shape of imidazole uncomparable.

Can you help me how to arrest the peak destorsion or any HPLC method for determination of Imidazole with good peak shape.

Not sure if I understand your message. If your peak shape is changing with the sample diluent, then you either are injecting too large a volume or your sample is dissolved in a solvent composition with a higher elution strength than your mobile phase or a combination of both. Try to make up your sample in mobile phase.

Can you disclose the mobile phase composition and column phase and make and analytical conditions , to throw more input to help you out.
I too accept the dissolution factor influences peak shape, but at the same time , suggest you to look in to the loading factor of column.
Please remember too many factors will influence the peak shape including your recorder / data processor speed. Is it your concern about the tailing factor etc. or only pictorial presentation ? How is the ascending and descending order of peak ?
