everybody, I am purifying this peptide called nesritide with RPHPLC ,the pI is 11.6,there is one intromolecular disulfide,and mw is 3462,the problem is that
1)I can not find the detection mobil phase on the pharmacopoeia,so I tried differen mobil phase ,I find TEAP is better than others ,so I use TEAP
2)there are several impurity peak before and after the major peak very closely,I have tried many mobil phase with different pH value, but I can not solve the problem ,we just have one pre-column from gracedac (jones chromatography,ID2cm)is there any parameters I can change ?can anyone help me
