I I am looking fo a Sep Pak cleanup for concentrating Vitamin D2 and D3 prior to HPLC analysis. Has anyone done this . The sample matrix is an extract after saponification of feed samples.into hexane

A Sep-Pak concentration of these very non-polar compounds from hexane might be tricky. One possibility might be with an alumina Sep-Pak. How much hexane do you have? I personally would rather consider an evaporation to dryness (hexane evaporates easily).
There is this 5-cm thick book on Sep-Pak applications. Do you have access to it? I do not have it at home, but I can check at work on Monday.

Hexane extracts after evaporation contain yellow residual from blood
serum. Vitamin D3/D2 and metabolites are reextracted into acetonitril
as injection solvent.

I have not found something specific for your application. However, there is a publication in J. Liqu. Chromatogr. 4(1) (1981) 155-164 on the analysis of the D vitamins from vtamin preparations such as resins, oils, concetrates and gelatin-protected beadlets.
