Hi. I'm trying to develop a RPLC method to quantify the hepatitis B surface antigen (HBsAg) in blood plasma but i don't know how to prepare the sample. I'm using the vaccine as the source of the HBsAg. For your information, the vaccine is a suspension (1ml) containing 20mcg of HBsAg, 500mcg aluminium hydroxide, 100mM phosphate buffer and 150mM sodium chloride.

From literature review, the HBsAg in yeast cell lysates were solubilized and reduced with a solution containing 4% SDS, 65% (v/v) mercaptoethanol and 1.3M DTT at 95 degree Celcius. But, since my HBsAg is in a suspension form, i don't whether the above sample prep is suitable. Also, is dilution of the vaccine possible and which diluent is recommeded?
