testimony on Imtakt Intrada Amino Acid column methods

Hello all,

I recently met with an Imtakt representative about using their Intrada amino acid columns for amino acid analysis. I was impressed and am considering giving their column a shot.

I do have two questions, however:

(1) Is anyone here analyzing a lot of samples using their columns? How has that worked out for you? What kind of elution are you using? What problems have you encountered? What is the column durability like (how many injections)?

(2) What is your sample prep to quantify free amino acids from biological fluids or fermentation broths?

I would be careful: The Eluent A contains 75 % THF, not good for capillaries and seals.

Hi Arne,

I've used AccQ.Tag previously for fermentation broth and mammalian cell culture media. Results from AccQ.Tag compared very well with classic AAA using ninhydrin and Dionex IPAD.

I used the older method with the NovaPak stationary phase, 4 micron particle size. These days the method is run much faster as I understand. Wasn't unusual to obtain 1500 - 2000 injections on the NovaPak column.

Thank you very much for the helpful replies! I had not been aware of the problems associated with THF use. I understand that methods using only water and ACN have also been developed on this system.

I have heard about the AccQ Tag method and that it is good but we have been concerned about adding a derivatization step. Do you use UV or MS detection for that.

Hi Arne,

I used fluorescence and UV detection in series for AccQ.Tag. I'd think that AccQ.Tag would be amenable to LC-MS, though at LC-MS I am honestly a beginner.
