Is it allowable for us to perform linearity test on our method validation Starting sample with unspiked standard solution? Thanks.
Hi jayroseville58

If you are talking of an assay you have to perform the linearity at 80 % ; 90 %; 100 %; 110 % and 120 % of the concentration you begin working when you develop the method.
In case you have to work near zero I think you have to find first the limit of quantitation (LOQ).

Best regards

Fernando
The merits and hazards of forcing your curve through zero have been discussed several times on this forum - use the search if you need more info.
Thanks,
