Is it allowable for us to perform linearity test on our method validation Starting sample with unspiked standard solution? Thanks.

If you are talking of an assay you have to perform the linearity at 80 % ; 90 %; 100 %; 110 % and 120 % of the concentration you begin working when you develop the method.
In case you have to work near zero I think you have to find first the limit of quantitation (LOQ).
