triglycerides analysis using gcmsd

Hi,

I had been using the GCMSD for FAME application. But recently, our company is interested to run the tryglycerides compounds in olive oil samples using GCMSD. But it seem like most of the journals or application are using GC-FID instead. Therefore, i would be grateful if anyone can any experience using GCMSD to quantify or qualify triglycerides?? Will they be any problem or do we need to perform derivatization prior injecting into GCMSD?? Thank you very much in advance.

You might find the bare metal in the detector a problem getting the peaks through the detector. Not a problem I have but it could happen. Sorry cannot help more.

We have used GCMS (HP 5890/5971) for intact triglycerides. We used a Quadrex stainless steel capillary designed for large compounds (10 meter, thin film) which can withstand higher GC oven temperatures better than polyimide-coated capillaries. The metal capillary was compatible with the 5971 MS, no problems. Normally we assay triglycerides after saponifying to fatty acid salts, making fatty acid methyl esters, then assay using a cyanopropylsilicone phase. Remember, assaying as the triglycerides will be tricky as there will be mixed esters (e.g. a molecule may have a palmitic, stearic, and oleic chain, and the positions may vary as well). We have also assayed intact triglycerides by HPLC using ELSD detector.
