Chondroitin Sulfate HPLC-UV analysis

Dear all,

I have the challenge to set up a methods for the analysis of chondroitin sulphate and glucosamine by HPLC-UV or HPLC-MS/MS... I know there are methods on the web that used IR and Light scattering detectors but I don't have them.
Any comment or suggestion will be much appreciate.

You can do the analisys of glucosamine by HPLC but the chondroitin is not possible.
Look at the USP 29 online from China.

You need to use ELSD for detection.

I found in greenmtlabs.com that by enzymatic hplc that treating chondroitin with chondroitinase ABC they produce disaccharides that can be monitored at 240 nm with rphplc and are very neatly separated.

Chondroitin is not UV active, in order to use UV you need to do derivatization or indirect detection. Any UV active impurity in the sample will mislead you.

Shouldn't each of these be detectable using positive mode MS due to the presence of amine functional groups? I suppose the separation could be difficult because of the large difference in MW but it could be worth a try.
