Poor Resolution/Broad Peaks

Hello,

I have been working on issues with a used agilent 1100 for a few weeks now.

My latest issue is a a problem with resolution and peak broadening.

I am currently trying to run a Bio-Rad gel filtration sample on a Tosoh TSKgel Bio Assist G3SWxl. My mobile phase is 0.1mol Na2So4 in 0.1mol Phosphate buffer (pH6.77). I have tried various flow rates, detection rates, injection speeds, and sample amounts to no avail.

Please, does anyone have any suggestions to get better resolution??

It'd be good to see chromatogram...
You can check flow path and all tubing. Maybe it is to long and too big diameter.
What detector you have in LC system?

A few questions regarding your set-up:

- What is the configuration of the 1100 HPLC you are running?
- Have you tried running a pressure test/leak test on the pump?
- Is the column compartment cover on?
- Is there larger diameter tubing out the end of the column to the detector?
- Is there any drift?
- What condition is the detectors flow cell in/what is the age of the flow cell?

- What state is your colmn in? New or first time use after 5 years storage?
- Is this a "real life" sample showing this effect, or is this a column test standard?
