Tablets: dependence of peak height/area on crushing or not

Hello,

I have a tablet with two well-distributed APIs.

When I dissolve the entire tablet as it is, I get higher heights/areas for API 1 than when I crush the tablet before dissolving

At the same time, I get lower heights/areas for API 2 for non-crushed tablets compared to the crushed ones.

Does anybody knows such phenomenons as well as a solution?

Sounds like you have both a solubility issue with one API and a sticking problem with the other.
I'd be inclined to find a LC-friendly solvent capable of fully dissolving both APIs in the tablet matrix and avoid crushing the tablets. Use a number of whole tablets and a larger vessel for your sample prep.

I'd do what DR suggests.

Why crush tablets if they dissolve? Extra step, chance of cross contamination, volatiles loss, etc.
