Do i need a special septum for SPME?
I ran our system with a normal Agilent septum and didn't have any problems. The only thing I did was pre-core them first by taking an old fiber and pushing it through the septum while it was installed in the inlet.

At my previous position is was common practice to precore the septa with an old fiber. Otherwise any reasonable grade sept with the dimple in the middle is advisable.
Do i need a special septum for SPME?
Yes you do. You have to use pre-drilled septum in your analytical equipment (for example in GC). Las time I "succeed" to break 3 of the fibers in 2 weeks just because my septum in the GC wasn't pre-drilled.
It depends upon how many samples your are running. Assuming your using a automated injector its best to use a merlin microseal for the long term, these are very realiable and we have very good reproducibility.
