Liquid chromatography capacity factor

Hi
At presently I have just joined analytical method development,
I am confused how to calculate capacity factor practically.

1. Measure the retention time of your peak and the dead time of your column.
2. Divide retention time by dead time and subtract 1

k' = (tR/t0) - 1

You don't calculate it, you measure it, just like you measure retention time. If you don't see a "solvent front" on your chromatogram, try injecting pure acetonitrile or methanol (I'm assuming you're using reversed phase here) and look for a small disturbance on the baseline.
