regarding auto sampler coller temperature
Discussions about HPLC, CE, TLC, SFC, and other "liquid phase" separation techniques.

for a RP-HPLC method can we have auto sampler coller temperature at 20°C and column oven temperature at 35°C. will this resulkt in any peak distortion.

Probably not, but the only way to know for sure is to try it.

I agree with Tom, I have run many related compound HPLC analyses that require the sample be held at 10C, and the column is kept at 40C to speed the analysis. haven't seen any distortion of the peak.

I agree too.
I set sample Temperature to 20°C and Column to 30°C without problem.
But I wonder if solvent temperature may disturb the run.
Can we see a different between 18°C solvent and 27°C if sample and column temperature are fixed (20 & 30°C)?

Ofcourse you can. Peak distortion would be highly unlikely.
Since the volume injected using the auto-sampler is relatively small (ususally few micro liters), it should not have any influence on the temperature equilibrium the column is already in.
Autosampler is usually kept at a lower temperature to supress any reactions going on in the sample.
