Has anyone experienced continuous problems on the PE FX-15 UHPLC. A brand new machine, one pulse dampener has been replaced... and others....
But right now I am running taurine std's pre-column derivatized by 1-fluoro-2,4 dinitrobenzene to form a UV active analyte. Mobile phase is 80/20 0.01 M phosphate buffer pH 6/ACN. Flow rate has varied from 1 to 2 ml/min from method opt.

The baseline is completely out of whack as you can see Image. What is going on. Thank you so much to everyone.

Hi Mike, I would suggest to increase the mixing volume. As your method is an isocratic method, you can double the mixing volume, maybe 1mL should be OK.
I did this and it worked perfectly.
