1. How dow set dwell time?
2. After automated compound optimization, can we directly do automated source gas optimization or else do we need to perform split infusion to optimize TEMP and Probe settings?
3. Can you please share your views @ outline of method development.

Hello Sri,

The dwell time is dependent of what you are doing. For a chromatogram you want 15 points measured per peak. So e.g. when the peak is 30 sec broad the dwell time is max 2 sec. This seems long however normally you have more traces so you have to divide for the number of traces. As well 30 sec is quite long nowadays.

The optimization for DP and FP you do normally out of a syringe with a flow of 5-10 ul/min. The optimization of the gas settings is done in a situation with LC running. You want to optimize the source conditions and in real this will be with LC with a higher flow (200-1000 ul/min). You can do this automatically but also manually is possible.

It is good to optimize the IonSpray voltage and temperature, the gases are less critical. setting 8 for nebulize, 8 for curtain and 3 for collision gas is OK in most situations.

Hello Sri

As far as I know, without experience the valve can be controlled by the pumps. In Analyst you can progran TE #1, TE#2 in the pumps tab. From this it should be possible.
