Dear,

Can i use HPLC grade solvent bottles (Amber/Transparent) as mobile phase bottles for the analysis, instead of buying Duran 1000mL bottles.
I am asking this because, some ppl say we can use, other say we should not use.

Regards

There are pros and cons to both, neither of which have any significant effect on the chromatography. I have used a full 4L solvent bottle of organic on HPLCs when I know I will consume the majority of the solvent. I don't move that bottle to another instrument to avoid cross-contamination due to the filters, which would have really no major effect on the solvent or chromatography, however I also don't want that solvent bottle then used for sample preparation.

I have used the 4L bottles for Methanol and Acetonitrile before with no problems. If you are running high flows on a continuous basis so that the solvent is used up each week then I believe you will be ok. The one you would need to be careful with is using a 4L for water. If using water you will want to empty and rinse at least once per week, wash with methanol or isopropanol to disinfect then wash again with water before reusing, otherwise you can have problems with microorganisms growing in you system.

The only other problem would be if your system pressurizes the reagent bottles, as these bottles are not rated for pressure and you will be using them at your own risk in this situation.

We've done similar with HPLC grade THF, but with 1 liter bottles.

Why not?

Yes

You may use Solvent Bottles for Mobile phase Use, But the Bottle Should be in transfer, Normally 99.9 % Solvent bottles in Amber Color, If we use Amber Color Solvent bottles as a HPLC Reservoir bottles we can not identifies Fungal Growth & Haziness of Mobile phase ,So don’t Use Solvent Bottles for Buffer Reservoir bottle,

Use Transferring & Standard Glass bottles for Buffer Reservoir bottle.

If buffer solutions are prepared fresh every day, amber glass bottles can be used without any problem. If you want to wait until the great white shark is in the bottle and you want to see him, other problems will popp up, for example clogging of the tubing, problems with valves, autosampler etc.!
