Sir,
Recently i was handled Morphine Sulphate Prolonged release Tablets project, Analytical Methods are taken as per British Pharmacopoeia, all methods are suitable for my formulation(Assay,RS) but problem with Content Uniformity by HPLC.
Tablets are not disintegrted in my diluent(Mobile phase) and try to dissolve in solvents combinations using Methanol and Acetonitrile but not usefull. My mobile phase containing 70:30:0.5 buffer: ACN:OPA.
Buffer:2.12g octane sufonic acid sodium salt.
I was used THF to break polymers(They are used PVA based polymers for formulation), it worked but Morphine sulphate degraded. Can anyone suggest polymer breaking combinations for my formulation.

Regards,
S.Ramakrishna.
I am solved my problem by using 0.2M mixed phosphtate buffer to pH 6.8.

Regards,
Ramakrishna.
