HPLC without extraction step ?

Hi,

Is it possible to perform HPLC based on a cell lysate without extraction step ?

What are the analytes? Lysate will be a mixture of all sorts of rubbish, at least use a guard column, but I would expect extraction is abetter way to go to protect your instrumentation, as a generalism
Where can I buy the kit they use in CSI?
I would like to quantify intracellular Talinolol (β1-selective beta blocker) in Caco-2 cell line. And I use a guard column.
