Negative spectra

Hello,

I have several samples measured using HPLC-DAD with a gradient. My problem is the reference wavelength I have chosen to zero the baseline might not be optimal, I've been getting negative spectra consistently. Is it possible to recover the original signal, without baseline substraction after the sample has been measured? or is it possible to change the reference wavelength after measurement?

I am using an Agilent chromatographer and ChemStation software.

Thank you.

Dear damt787,

I have looked for the same feature and have not found it. When I've run into the same problem I recollect with the reference off & using a wide range on my UV acquisition (190-600nm+), check the UV spectra of my peak, and evaluate whether to use a different reference or keep it off based on what the spectra tells me.

Sincerely,
Keith
