Hi everyone!
I validate a method with Vitamin B12 as active ingredient and facing a problem with accuracy.
It have small concentration (about 0.01%) and the best recovery i have using external standard method is 50%. Standard addition proved doesn't give good accuracy and repeatability result.
To overcome this problem i start using standard added with plasebo. With this method i have about 94%-98% recovery, but linearity from this standard is less than 0.99.
My question is :
1. Can i using this kind of standard preparation? (my concern here is this kind of standard preparation gives area under curve less than it should be compared with if i don't add plasebo)
2. Is it ok to change linearity acceptance?
3. Do you have any other suggestion to overcome this problem?
