carbohydrates problems

We did analysis of glucose syrup (from starch hydrolysis) while ago and we used conditions recommended for our column (amino) and detector (UV). We had god separation and quantitation, however we had one unidentified peak between RT of glucose and maltose. We tried some other monosaccharide and disaccharides (galactose, sorbitol even HMF (one of the degradation products of glucose)) and we didn’t get any match. We also tried GC/MS identification with appropriate derivatization but our library isn’t really the best for sugars. Did any one had similar problem?

Hi,
Can sugars be done successfully using a UV detector? I do a lot of work with fermentation sugars and other carbohydrates and I thought that they could only be picked up using RI detector..
Please let me know ..

Hello,
RI detector is indeed most used detector for sugars, however we found several different official methods for sugars analysis with UV detector( for example AOAC, Agilent Applications etc.) So it’s safe to say that you can use UV detector. My experience with this detector is very good. We had no problems what so ever. LOQ and sensitivity were excellent, as well as repeatability. We analyzed several different sugars from monosaccharides to tetrasaccharides and the results were equally good. If you need any more information please let me know.
