I will analyze stereo isomers in bortezomib lyphilyzed cake 3.5mg/vial(for injection).I was developed a normal phase method by using chiral pak AD-H Column and mobile phase contains 920:40:40,N-hexane:ethanol:IPA.For API This method is no problem,only problem with lyphilyzed cake,its having 35mg mannitol.

I am using mobile phase as diluent,but mannitol not dissolved in n-hexane,ethanol and IPA also.It is only dissolved in water or dmso, these are not misciable with my mobile phase.If i use pure DMSO.Ethanol or IPA peak shapes are effected what i will do help me?

Mannitol is very polar with a Log P more negative even than glucose (-3.262 vs -2.49 ref. ACD/Labs). It will definitely need a more polar solution than what you are using. With 92% hexane, I think you may be S.O.L with Solubilizing this sugar alcohol. You maybe could play around with dimethylacetamide instead of DMSO, but I still don't think it will work with these conditions.

What about dissolving in water and then doing a liquid/liquid extraction with organic?
