LC-MS/MS new method development --Help requested

Hi,
I am new to LC-MS. We recently bought a used API-2000. We got it tuned by a service engineer. Now, I want to develop a method for a compound of known molecular weight, to get used to the instrument. However, I have couple of questions.

Should I start a Q1 MS scan first to get the Q1 Ion Mass and then to MRM scan to get Q3 ion? or

Directly start with MRM scan? If so what will be Q1 Mass and Q3 Mass?

I kindly request all to help me with steps to develop a method successfully. I have done one analysis using a previously developed method but not comfortable. As I want to understand the principle and actual way to work on the instrument.

And FYI, it's a 6430, not a 6340, and it's a triple quad, not an ion trap.
