Varian Galaxie - fraction collection inconsistency

Hi,

I'm new to this forum, so apologies if I am posting in the wrong thread/manner.

The fraction collection window was set from 12:00-16:00 in my method, and I had set it to collect at 240-second fraction in each tube. However, Galaxie decides to collect a fraction from 0:06-12:52 of the run, and everything after that goes to waste. Any ideas what went wrong?

Hello,

You should contact Agilent for technical support, as it will be difficult to help you this way. Someone needs to check the system you are using (which modules, what driver's versions, what Galaxie version), and check the method you set.

Agilent now owns Galaxie. Be careful, as a lot of people in Agilent think Galaxie is a Persistent Systems product now, whereas it is wrong. (Persistent bought the office where Galaxie has been developed, but not their products)
If you have a SMA, they have to help you.

Maybe, a little mor information can be helpful:
What are the parameters inside the Time window / the configuration of the fraction collector itsself.
Whitch Type of fraction collector are you using? ( 701 or 440?)

Hi all, thanks for the replies.

While I am uncertain of the configurations that you mentioned, it is a 701 fraction collector. It is set to collect in a serpentine manner (non-peaks go to valve), and moves to the next tube for each fraction.

I recommend the following parameters, parameters, I dont write about should be unchecked:

a)Collector parameters:
Mode=Time
Pattern=serpentine
Flow delay= 0 (depending on your Tubing&flowrate)

b)Fraction collector Mode:
check only Time here (if you want to collect by time),
In the Fraction windows box, you can enter your time window (12-16min)

c) racks:
enter your Rack
Fraction size = 240s

Does the 'flow delay' parameter help adjust for dead volume?
the fraction collector is an isco OEM for varian
how have you connected it to the software?
is it with direct control of the software via RS-232?
maybe it is best to have it working on a closure contact mode sent from the pump and have the method run by the fraction collector itself

Yes, the flow delay ( in seconds) compensates the dead volume ( the volume of the tubing between the flowcell of the detector and the valve of the fraction collector)
The flow delay is the tubing volume divided by the flowrate

The Galaxie software programs the same parameters that are programmable by the planel of the collector - so the way unmgvar recommends is also a way that should work well.
