Desperate PhD student - New to Chromeleon and failing!

Hello!

So, as the subject says, I am desperate. I have been chucked in at the deep end with a poorly maintained Dionex HPLC system and no knowledge of Chromeleon. I have so far succeeded in cleaning out the system and getting sensible separation of the monosaccharides I am trying to analyse... It only took 5 months of my project (battling with finance to get funding for parts etc!).

My downfall is with Chromeleon. I was left a qnt. and pgm. file that I just needed to tweek a bit and that should have been it. But I think I'm missing a major component on the report files - the amount in mg/ml. It just says n/a... agh! Clearly I need this to be able to know how much of each suagr is there but no matter what I try (including following the Chromeleon tutorial and help) I can't seem to get figures in there!

I really am at the end of my thether with this and if anyone can please give me some advice or know how to fix this please please let me know!

You need to make calibration curve.

Find "calibration" in help file.

As a work-around, put the peak areas and your calibration data into Excel and do the number crunching there.

If you look in the short report (icon looks like a white table with blue header) and the amount says NA then Chromeleon can't quantify your results. Check the following;
  • Is the peak named and assigned correctly
    Do you have Calibration samples in your sequence with type = calibration
    In the method (qnt) file is the calibration method set (prob total)
    In the Amount tab do you have amoutn columns created for your standards
    Are those Amount columns filled in with amounts
    Can you see a calibration curve if you click the Calibration icon (looks like a cal graph)

I'm working with Chromeleon analyzing Glucose, Fructose, Saccarose, Lactose and maltose. Perhaps I can send you Bckup of my Chromeleon files...Then you can modify them at your convinience
Do you feel abble to handle the backup?

Sorry, no. The system I have is a work one, so not possible.

You should be able to do this via the help, just go through the method setup section.
