Hi all,
some one can tel me why we use aceton 0.05% for HPLC gradient autovalidation? (I am using shimatzu HPLC).
and can I substitute water by MeOH (I have some dificulties to pass the test using aceton 0.05%/water).
thank you.

Acetone is unretained, and it has a UV signal.

If "gradient autovalidation" is a test of pump and gradient proportioning valves then fact that acetone is unretained may be neglected, because test is executed with pressure restrictor instead of column.
