Separation-polysaccharide from polysac-protein conjugate

Hello everyone,

My question is related to a step in our sample preparation for estimation of % of polysaccharide fragments released (on depolymerization) from polysaccharide-protein conjugate using Dionex IC 5000.
In this step, we are using SPE C4 cartridges (Particle size: 13 um, Pore size: 300 A, source: Grace Vydac) to separate the polysaccharide from a mixture of protein-polysaccharide conjugate and polysaccharide fragments. It is well characterized that these cartridges efficiently retain small protein/peptide molecules from a mixture passed through them. However, can the cartridges retain the large protein-polysaccharide conjugates with such high efficiencies ? Or could you please suggest any other alternative separation technique to achieve efficient separation of polysaccharide fragments from the conjugates?
