column washing

Hi,

I'm using a small silica-NH2 column where I immobilized a proteoglycan. I inject a protein solubized in PBS but sometimes I have like a clog in my column.
Normally I wash it with 10% MeOH.
Do you know if it's the best way to wash it or do you use another solvent?? I'm affraid of damaging my column with my immobilization.

The general rule is to flush with something that is a good solvent for whatever gunk is in there. If the 10% MeOH is working, I would stick with that rather than risk killing your proteoglycan.
