amino acid analyses with BSTFA

Hi !

I'm new in this forum but I use to read the posts.
Today I would like to ask a question. I need to analyse an amino acid which the structure is near from glutamine in a plant extract.

I can elute it on the Zic pHILIC from SEQUANT with a retention time of 12 minutes, but in my plant extract I have another compound with a very near structure, and the two coelute.

So I would like to try in GC but I don't have a lot of experience in GC . I read that the amino acid can be derivatized with BSTFA in general doc on GC. I have also a DB 5 column.
Does anyone know conditions for the derivatisation ?
Which solvent compatible with BSTFA is used to dissolve the amino acid?

Hello Mel, I have one procedure for amino acids derivatization. with HFBA
if you have your hidrolysis completed, you would have to:

Take 0.2ml of yuor sample ( or the equivalent to 1mg of protein)
Dry under N2 at 45 degrees
Ad 100ul of 3M AOcCl/isoButanol
incubete 20min at 120C
Dry under N2 at 45C
at HFBA 100ul
incubate againt and dry
disolve with Ethyl acetate.
and you should ready to inject.
