Hello together,

could someone please tell me how to determine system dispersion the easiest way?

Hi Anja,

have a look at a column "Care and Use" manual from Waters (e.g. XBridge Columns, Docu: 720001366EN). There it is such a procedure described
(look also for band broadening/band spreading etc.)

In general:
- replace column with union
(- add some pressure device after detector (or between pump and Injector))
- set the data rate as high as possible
- use a low flow rate (e.g. 0.5 ml or below)
- inject a small volume (1-5µL) of a test mix of enough concentration as to get enough absorbance (e.g. coffein, uracil, parabens or similar compounds)
- evaluate the peak width at 4.4%
