Waters HPLC system

Hi

I am using a Waters 2489 UV/Vis detector and been having a lot of problems with it - mainly inconsistencies in separation if any at all. Also, their Autosampler 2707 doesn't seem to allow me to make more than 50uL injections although it has a 100uL sample loop.

Has anyone been having similar problems with Waters?

Check the settings in your method to see if you are set to use a full loop or partial loop. Oftentimes if the method in the software is set to partial loop on a 100uL loop, then it will allow 50uL as the max injection, but if you change it to full loop you can do the 100uL injection.

Also, the detector has nothing to do with inconsistencies in the separation. It merely sees what's coming out of the column.

the 2707 is an OEM from spark holland
http://www.sparkholland.com/products/au ... ers/aliast

this sampler is a partial or full loop sampler
so you can inject up to 50ul or a 100ul
you can probably inject also in between but due to the design of the sampler the RSD will not be great
