How critical is the effect of the TFA concentration on the retention time and separation of peptides on a reversed phase column? Will you see a difference when the amount of TFA is 0.1% instead of 0.2%? And if so will you also see a difference when the amount of TFA is 0.12% instead of 0.1%?

How much (if any) difference you will see depends on your peptides. Among other things, TFA acts as a weak ion-pairing reagent, so increasing the TFA concentration will increase retention of more basic peptides.
