I can not find anything to explain to me thoroughly how to calculate the concentration of a substance (example vitamin) by HPLC with external standard technique!!
That is, once a calibration line prepared the compounds of my .... how do I know the concentration of alpha tocopherol, for example., 2g extracted from a food matrix with 12 mL of hexane, taken to dryness under nitrogen and 1 ml reconstituted in the mobile phase 200microL!!
I enter the area in the equation of the line, I calculate the concentration, multiply the volume of injection (10microL )..... and then ???????? multiply by the dilution factor? and how would it be? I miss the steps !!!!!! Thanks for your support!
