SPE oil extraction

i want to extract a Pharmaceuticals from olive oil samples, i don't know which kind of solvent can i use in this extraction

Hi mmajdop,

Thought I'll answer your question here. Depending upon the solubility of the pharmaceutical you are trying to extract from olive oil. Methanol is mostly immiscible with oil, but if you prepare 10 to 50% water with methanol, you'll get near complete immiscibility with oil. On the other hand, acetonitrile is an excellent solvent to extract from oil. You just need to check the solubility and polarity of the pharmaceutical. Hope this works for you.

thank you all

i will trying to dissolve a pharmaceutical standards in (MeOH:H2O), adding the standards to the oil, homogenize, passing the oil sample with a standard in a SPE C18, and Elute the Compounds with acetonitrile .

Remember to think about the pH and the pKa of your compound; if your analyte are polar you could use a mix of Waters:MeOH (or MeCN) at low pH in order to extract basic compounds and high pH for acidic compound followed of hexane clean up for remotion of more polar coextractive compounds. Nevertheless; remember if you use high concentration of Orgnic solvent the recovery of your analytes could be low depending of what SPE cartideg you use. If you can extract your analytes by pH this is going to be much selective and I think the main objectvie of SPE it would br to concentrate your sample.

I would strongly second oscarBal's suggestion if you can do it this way. Convert it to the ion, extract it with water, convert it back and extract it with solvent. If do-able, very effective.

Depending on the size of the analyte, you might consider GPC as a clean-up.

what do you thinks is more effective C8 or C18 SPE for the samples extraction??
and whitch detector is More sensitive for the determination??

i thinks HPLC is beter than GC for the determination of this samples??

C8 will provide you with a cleaner extract for a highly hydrophobic and uncharged analyte. If only slightly hydrophobic, use a C18. C18 is less selective. Another consideration is endcapped vs unendcapped. If your analyte is highly hydropbobic use endcapped. If your analyte has polar functionalities and is weakly hydrophobic, go for unendcapped.
