hi all,
I am a fairly to the Liquid chromatography world so please bare with me. I have a Shimadzu analyical system that I am running with a polymer column. Recently, I received a high pressure warning and the column started leaking. i have been trying to unclog the column but that is not working. It is currently running at 4300 psi and it fluctation about + or - 40 psi when I am pumping at 0.5 mL/min. I then bypassed the column and the pressure drops to 75psi but still fluctuations like I stated earlier. I thing that the system was running without the deagasser on for few hours which could be part of the problem. Also, I had used a buffer that may have precipitated on to the column. What can I do to save my column?? Please help!!!
First of all please disconnect your column from the detector, than start to flush your column by reversed flow with 0,1ml/min at first. If buffer was the problem. Than disconnect your column from the system and flush the system with high flow rate to eliminate possible air bubbles from the system. Good luck.
