I was just reading about TCEP as a better reducing agent than DTT or B-ME. I read that it can be used with IMAC and is more resistant to air oxidation.

Does anyone have hands-on experience with TCEP? What molar ratios to use with my protein? Can I use it with IEX resins?

Hi Joey,

I have used TCEP on the Beckman Coulter PF2D system and it works well for raw proteome analysis at 5 mM concentration. Beckman usually used this with the Chromatofocusing columns they employed on this system. It was a good gentle reducing agent and does work well with IMAC. The company who has the columns still for this as well as oth IMAC columns is Eprogen and you can find them on The web at www.Eprogen.com
