I am using Methanol as a diluent with 10000ppm concentration, (Compound is Acidic in nature) but peak shape is not proper even after increasing the buffer strength of mobile phase ...So Please let me know that How can I get Sharp peak?

As seems to often happens, you have provided almost zero information that would be usefull in solving your issue. However, based on the limited amount of information provided it is likely one of 3 issues (or a combination of these).

1. Sample to concentrated (10000 ppm??)
2. Sample Diluent to strong (100% methanol)
3. Injection volume too large

Of course it could also be one of 10 (or more) other common problems that cause pook peak shape.

In nature many more acidic substancies - gastric HCL in stomach , citric acid in lemone, free thriphospate ATF in sperm... 8)
