Peptide detection

Hi colleagues,

I am going to carry out the detection of a peptide. It has different ionization sites due to have several amino groups and hydroxyl groups. My question is...I have to add TFA in the mobil phase to aid protonation...or with ACN / H2O could be enough?

TFA is used as an ion pairing agent in peptide separation, but it isn't a requirement for every analysis. You could start with ACN and 0.1% formic acid and see if that works for you.
