Alternative to Glycine buffer (pH 9.0)

Hi all,

I'm searching for glycine buffer alternative for mobile phase.
I'm using gradient method : sol.A Glycine buffer (pH 9.0), sol.B : Acetonityl:MeOH 85:15.

I'm searching appropriate buffer at pH 9.0 for UPLC system (but not Ammonium based).

Please help :P

Does it need to be volatile? If not, how about Tris?

No, it doesn't have to be volatile.

What about phosphate buffer, prepared from tri-basic and di basic phosphate?

It should work as the pka's are 7.2 and 12.32, barely within the +/- 2 "rule".

Another organic buffer is CAPSO, pKa 9,7.
Or 1-Methyl-piperidine
Or Borate, but it is not volatile.

Borate buffer -good stabile buffer , but in LC-MS borate buffer must not use.

Nasty (Ungly) it is.
Volatile, malodorous, unstable and RP-silica gel "eats". But because of volatile buffers for UPLS-MS the ammonium is most volatile.

2 solntzevorot (Midsummer)=> by nick can be seen from the "brozer slavjans". Come on (part of) on anchem.ru

Very likely when you worked with the ammonia buffer and know that "it's not sugar". :D

The buffer no need to be volatile.

Whith ammonium buffers (at least with those I have in the lab) I experienced with to much system peaks in the UPLC system.

to DSP007 : thanks for the link, I freely speak Russian and Ukrainian. (Born in USSR :) )

You shouldn't experience system peaks with ammonium buffers. If you saw peaks is probably either the low purity or contaminated salt. I would try again with some new high purity container.
