docetaxel extraction issues

Hello. I am having difficulty extracting docetaxel from mouse tissue that has been treated with docetaxel. I have read several protocols using acetonitrile solvent to homogenize the organ and then using the supernatant (solvent) in the subsequent steps for analysis. However, my samples of homogenized tissue in acetonitrile do not demonstrate the necessary docetaxel peak.

I have conducted control studies with just docetaxel and paclitaxel alone and I have seen the peaks (indicating it is not an instrument issue). I have also taken blank tissue (not treated with docetaxel drug), homogenized it in acetonitrile, used paclitaxel for internal control and upon HPLC analysis saw a weaker paclitaxel peak than compared to free paclitaxel (as mentioned above). This leads me to believe that the acetonitrile solvent is not extracting the drug from the tissue.

Any suggestions please???

Soluble in ethanol, methanole, chloroform. Chlooroform extaction is standart method in toxicoligy.

Do you have direct experience with this issue? With docetaxel and extraction?

How about with tetrahydrofuran? Would that work as well?
