Inositol

Hi Everyone,

I'm new to this forum, and hoping someone can help me out. Sorry if my terminology on the subject isn't up to scratch, I've only just started my project.

Has anyone had any luck using reverse HPLC with inositol? At the moment i'm finding my standard solution is going straight through with my mobile phase and I don't have any retention. I'm working with a 0.0001M standard. Initially, I tried a methanol and water mobile phase with no luck. I've tried changing it and using an ion pairing reagent however I'm still having no retention. I'm using a C18 column currently (although am thinking I may need to use an anion exchange column). I've been using a UV detector, however have access to an ELSD detector.

If anyone has any suggestions (or needs clarification of details of my experiment) or they've had success I'd love to hear them :cyclops:

For Inositol I would use a HILIC column ( Hydrophilic Interaction). If you have a Diol or NH2 column in your lab, try them first. ACN 75% and Water 25% would be a good starting point for your mobile phase. Otherwise ask manufacturers of HILIC columns for a free of charge test column. Good luck.

As Gerhard pointe out using HILIC it is simple to get retention on Inositol see application note below using ELSD.
