heparan sulfate analysis

Hi everybody,

I'm new on this forum, so please be indulgent with me!!!
I'm trying to analyse heparan sulfate proteoglycan interactions with proteins.
I would like to use HPLC with a spacer between silica and heparan sulfate proteoglycan.
Does somebody know the best spacer I can use it for?
And if somebody has tested it, did he had problem with fixation ?

Between 4-6 carbons are commonly used. How do you plan to bond it to the silica, i.e. what will be the reactive group?

I want to study both heparan sulfate et heparan sulfate proteoglycan.
For heparan sulfate I want to bind the aldehyde group, I found a publication from BJORKLUND et al,( J of Chromatography A, 728 (1996) 149-169 where they did it, but they did it "in batch" and I would like to realize it "in situ".
For HSPG, I think I want to try to fix aminoterminal end with glutaraldehyde.
