Hello I am trying to analyze the amount of amidoamine in cocoamidopropyl betaine. I am using an ion exchange column and the mobile phase has an ion pairing agent (TFA). If I use my mobile phase as my diluent will my active ingredient be separated from the solution prior to injection? Although cocoamidopropyl betaine is soluble in 90:10:0.1;Water:ACN:TFA my amidoamine is not soluble and separates in volumetric flask depending on the concentration. As the ACN increases both becomes more soluble. Does the analyte of interest have to be soluble in the mobile phase to obtain good separation? It is soluble in 50% ACN in water, but my peak of interest is very broad in the sample, but is acceptable in the standard. Any suggestions in regards to this matter would be greatly appreciated.
