Sebum analysis by HPLC [June 15, 2004]

I am a beginner. I am trying to analyze the various components of sebum by HPLC. I am looking at help on what column/mobile phase and methods would be best.

Sorry for the delay in responding. Sebum is the stuff that is secreted by sebaceous glands in the skin and is responsible for the oily feeling on the skin. It is a mixture of fatty acids, triglycerides,
cholestrol, squalene, wax esters etc.
I am looking for a method that can distinguish between the classes of compounds mnentioned above. If individual fatty acids etc. can be resolved it would be nice but is not absolutely necessary. Thanks again for your help.

Try TLC first (check lipids in any book on TLC).

It can be done with HPLC, but the problem will be a suitable detection technique. What is your detector?

thanks for the responses. I have access to a UV and RI detector. In addition a light scattering and FID detrector could be arranged if necessary.

Which dtector system do you advise?

I am trying to study oxidation products of sebum. The stuff that causes yellowing of soiled garments over time. The high molecular weight compunds produced are difficult to analyze by GC. Polar molecules also pose a problem with GC. I was hoping to use HPLC to complement my GC analysis. Fill in the 'holes' so to speak.

Looks like you found your life´s job. If those substances are yellow and can be dissolved then a UV/VIS detector will do. How do you get this stuff off the cloth?
Still, seems like a typical exploratory job for TLC.
