Is it possible to determine the structure of a unknown protein using Electron spray ionisation or it is possible only with MALDI

Primary structure refers to the sequence. Prior to MS you have to digest your protein. To analyze peptides after digestion you can use both ESI or MALDI but for small peptides ESI or matrix-free MALDI would be better.
Secondary structure: CD, FTIR
Tertiary and quaternary structure: fluorescence spectroscopy, NMR, X-ray
