Mivacurium consists of 3 izomers: cc, ct, tt. The most important is additon of esterase inhibitor to blood immediately after sampling or to plasma.

Three isomers are not available. You can try separate their on HPLC from
injection preparate.

The best column is Supelco Discovery RP-Amide C16 150x4.6. pH<3 ion-pair
Detection spectrofluorimetric or Varian Prostar UV detector.
Extraction - solid-phase.
2 articles exist in literature. IS available from producent or verapamil.

Pharmacokinetic study is not easy because very short half-life of active izomers and periferial elimination. Concentration-independent model is sugested.

Is there any way you can provide us with a sample. We will do free method development for you. You do not need to use ion-pairing reagent for this one. My gyes is that peak shape is very poor on traditional RP columns due to the presence of two quarternary amines, but it is not the case for Primesep. If we get a sample we can do a screening in a few days. No obligation to buy a column from us.

I’ve already bought a Whatman Partsil-5, 4.6 x 250mm silica column. I think silica columns are good at separating structural isomers. I’m going to try this column first. If I cannot get a good separation, I will send my sample to SIELC-TECH to try on your columns (but I need to ask to my boss first ) or try the column that Kazimierz suggested.
