column very high back pressure

Hi All,

I amMr Khalid Rauf, a PhD scholar working on neurotransmitters, in distinct rat brain tissues, like, frontal cortex,striatum,etc using HPLC, with coulchem III, detector using MD-150 column.i use phosphate buffer 90:10 ACN, ph 3 and 0.6 ml flow rate.I experience more than3500psi pressure at 0.6 ml.How ca i clean my column .

You didn't specify the diameter. While I've never worked with that specific column, 3500 psi for 0.6 mL/min through a 150 x 3mm column sounds a bit high, but not unreasonable. If you're using the 1m ID column 3500 psi would be quite low.

First check to make sure there is no blockage elsewhere in the system. Then check with Dionex (ESA) confirm that back-flushing the column is OK. If so, then turn the column around and run it in reverse for a half-hour or so.
